Major transcriptomic, epigenetic and metabolic changes underlie the pluripotency continuum in the rabbit

Since 2008, researchers from the Biology of Reproduction, Environment, Epigenetics and Development Unit (UMR1198 BREED) have been collaborating with the INSEM unit (U1208,INRAE USC1361 Stem Cell and Brain Research Institute) to characterize pluripotency in the rabbit embryo and to derive naive pluripotent stem cells (ESC or iPSC) in this species. This long-term collaboration has already resulted in several publications and has just reached a new stage with the publication of an article in Development.

In order to identify key markers of naive pluripotency in the rabbit, we performed a double transcriptome analysis of embryos (bulk and single cell RNAseq) complemented by molecular analyses of the metabolism and epigenome. These data allow us to better understand the processes regulating the pluripotency continuum in the rabbit embryo as well as the segregation of early embryonic lineages. The results show that the pluripotency continuum in rabbits is more similar to that of primates, while that of mice has many particularities.

This information also offers new avenues for the study of pluripotency in vitro.
