The LRR-RLK gene LaCCR1 controls cluster root formation in white lupin

Root systems display a remarkable aptitude to developmental plasticity. An extreme example is the production of Cluster Roots, which are dense clusters of short lateral roots, as a response to low phosphorus. My lab has developed white lupin as a model plant to study these structures, sequencing its genome (Hufnagel et al., 2020a) and pangenome (Hufnagel et al., 2020b) and generating descriptive data regarding cluster root development (Gallardo et al., 2019; 2020). In parallel, we identified mutants altered in cluster root formation; the constitutive cluster root (ccr) mutants produce cluster roots in repressive conditions (high phosphate). By sequencing 4 alleles of ccr1, we identified an LRR-RLK gene as bearing the causal mutation. This protein acts as part of a long distance mechanism to repress cluster root formation, in a manner that is reminiscent to the Autoregulation of Nodulation in Legumes for the control of nodule development. We are now trying to decipher the molecular pathway associated with the CCR1 kinase and trying to understand how it regulates organ development in response to the phosphate status. I will present our latest data on the ccr mutants and hope to get feedback from the community.
