Plants are grown for 5 weeks in a growth chamber. Leaves are inoculated with a bacterial suspension( 10*7 cfu/ml) using a syringe.
Jiffy pellets grown Madicago truncatula plants are cut at their bottom third and soaked in a bacterial suspension at 10*8 cfu/ml, wilting is observed around 5 days after inoculation and the whole bioassay is completed around 15-20 days after inoculation. In vitro grown Medicago truncatula are inoculated directly on the exposed roots with 200µl/plant of a bacterail suspension at 10*8 cfu/ml, wilting can be observed after 10 days, the assays is considered finished at 21 days post inoculation. the maximum of bacterial colonization is usually obersed at around 10 days post-inoculation.
Tomato plants grown in the greenhouse are watered with 50ml of a bacterial suspension at 10*8 cfu/ml. there is no need to artificially wound the plants, the disease (wilting) starts around 3 days after the inoculation and the bioassay is finished in 10 to 14 days.
plants grown in jiffy pellets have their roots cut at the bottom third and soaked with a bacterial suspension at 10*8 cfu/ml. The disease (wilting) appears around 4 days after inoculaion, the whole bioassays is usually finished in 10 days.
